Common Methodologies for Peptide Research

This document provides standardized protocols for common peptide research applications. These protocols serve as starting points and should be optimized for your specific research needs.

1Standard Reconstitution Protocol

  1. 1Remove peptide from freezer and allow to reach room temperature (15-20 min)
  2. 2Clean vial stopper with 70% isopropyl alcohol, allow to dry
  3. 3Calculate required solvent volume based on desired concentration
  4. 4Using sterile syringe, slowly add solvent along vial wall
  5. 5Gently swirl until fully dissolved (do not vortex)
  6. 6Aliquot into sterile tubes if multiple uses planned
  7. 7Label with date, concentration, and storage conditions

2In Vitro Cell Culture Protocol

  1. 1Prepare stock solution at 1mM in appropriate solvent
  2. 2Filter sterilize through 0.22μm filter if needed
  3. 3Prepare working dilutions in cell culture medium
  4. 4Add to cells at desired final concentration
  5. 5Include vehicle-only control wells
  6. 6Incubate for specified treatment duration
  7. 7Assess endpoints as per experimental design

3Stability Testing Protocol

  1. 1Prepare peptide solutions at standard concentration
  2. 2Aliquot into multiple identical containers
  3. 3Store under different conditions (4°C, -20°C, RT)
  4. 4At each timepoint, analyze via HPLC or LC-MS
  5. 5Compare to time-zero reference standard
  6. 6Document any degradation products observed
  7. 7Calculate % remaining activity over time

General Best Practices

Always work in a clean, dedicated space
Use sterile, low-binding plasticware
Minimize exposure to light for sensitive peptides
Document all procedures and observations
Include appropriate controls in all experiments
Validate peptide activity before large-scale studies
